Steelbacks captain David Sales was to the fore, smashing 78 from 42 balls with seven fours and four sixes.
He shared an unbroken 82-run sixth wicket stand with Damien Wright, whose 38 occupied 20 deliveries.
Aussie Wright took 3-17 and Ben Phillips 4-28 to bowl the Gladiators out for 143 in reply, Mark Hardinges top-scoring with 40.
The Northants total was three more than the previous best, first achieved by Gloucestershire in 2003 and equalled by both Surrey and Leicestershire last season.
