Nottinghamshire, electing to bat first, put up a good score of 179-9 with veteran Mark Ealham smashing six sixes in hitting 45 off 17 balls.
Gareth Breese did well with the ball, taking 4-21 in just two overs.
Durham's reply began well with Nathan Astle's 64, but 20 runs were needed off the final over, and Gordon Muchall and Dale Benkenstein did enough.
Muchall finished unbeaten on 64, while Benkenstein hit a six and a four in 13 from four balls.
The winning hit came with three balls remaining, for Durham's first win this season in the Twenty20.
