Leicestershire will face Glamorgan in the second semi-final before the final takes place on the same evening on Saturday, August 7. A reserve day has been allocated on Sunday, August 8.
Surrey, who beat Warwickshire in last year's final at Trent Bridge, are yet to lose a game in the two-year history of the competition.
The Lions booked their place in the semi-finals with a 14-run win over Worcestershire on Monday night.
Lancashire, the only one of the four sides not to have topped their group, joined them by crushing Hampshire by nine wickets at Rose Bowl.
Lions batsman Mark Ramprakash is expecting a tough semi-final and hopes his side's experience in the competition will work to their advantage.
The former England batsman told Sky Sports: "It's obviously a big game and there will be a great atmosphere.
"Both teams will hope to have all their big players available. That will make it a bigger occasion.
"I think having been through 11 or 12 games unbeaten, a lot of our players have experience of when things are not going our way to keep calm and focused and that really helps."
Lancashire all-rounder Dominic Cork is under no illusions as to the size of the task.
Cork said: "Obviously they're the champions and it will be a difficult match but we have got to beat good sides to win the trophy and it will be an exciting first game.
"They've got the trophy from last year, they're unbeaten this year and they'll be the favourites."
The first semi-final will take place at 11.30am before the second gets under way at 3pm. They will then be a concert featuring Liberty X and Natasha Bedingfield before the final starts at 7.15pm.
Glamorgan skipper Robert Croft is confident the Dragons can go on and win the competition.
Croft, whose side beat Warwickshire in the quarter-finals, said: "We will play anybody at the moment. Leicestershire have found a format that has been successful for them.
"They are a fine side and it is up to us to put in a performance that is worthy of winning."
Leicestershire were beaten at the semi-final stage last year but Foxes director of cricket James Whitaker believes his side can go better this time.
Whitaker said: "We would take whoever came along. We're pretty comfortable with Glamorgan.
"It will be a great day, it's been a good competition so far and the lads are looking forward to a good day."
